I have a standard USA strat single coil pickups etc. I want to increase the sustain a la SRV. I've tried compressor effects (via a Pandora PX4) and .11 & .12 e string but it still sounds a little 'weedy'.

I don't want to mess around with effects too much and lose the unique Strat chop.

What are my options ?

is you trem floating? if it is, try blocking it, or put some more strings in the back.

He also tuned down a semitone (hence playing some stuff in Eb) and had bass guitar frets fitted to increase sustain.

You could try raising the pickup height a bit. Not too much or you can get wolfnotes and the like.

This is what you are looking for. http://www.victorious.co.uk/theguitar/index.html I don't know if you would find one... or what it would cost. last I heard, the guy who built them wont build anymore. And Mine is NOT for sale. but controlled and consistent sustain and Strat scale length.
